Pharmacokinetics of ketoconazole in normal subjects

The serum pharmacokinetics of ketoconazole were studied in six volunteers. Each received 200 mg and 400 mg of ketoconazole as single oral doses on two occasions separated by 2 weeks. Serum ketoconazole concentrations were measured over a 48 h period using a microbiological agar diffusion method. Peak serum concentrations were proportional to dose and occurred within 3 h. The mean serum after the 200 mg dose was 122 ± 25 mm (mean ± s.o.) and after the 400 mg dose was 160 ± 29 mm. These differences were significant (P <0.01).
