The use of ferret trachea organ cultures for therapeutic studies of anti-influenzal drugs
II. Comparison of four compounds in tissue culture and in organ culture

The activities of spiroamantadine, cyclooctylamine, virazole and amantadine against influenza A/England/42/72 have been compared in therapeutic experiments in tissue culture and in ferret trachea organ culture. Measurement of virus yields from tissue cultures treated 3 h or 24 h after virus inoculation showed differences between the compounds which were not revealed by simple titration experiments. When the four compounds were used for treatment of ferret trachea organ cultures 24, 30, 48 or 72 h after infection spiroamantadine and cyclooctylamine showed similar activity to that of amantadine at equivalent molar concentration but virazole was more active than the other three compounds. The results also suggested that drug-resistant virus may sometimes develop in cultures treated with cyclooctylamine.
